Notes Antigen Retrieval Solution is a Tris/EDTA, pH 9.0, intended for Heat Induced Epitope Retrieval (HIER) in tissue sections prior to adding primary antibodies. This solution is designed for unmasking a wide variety of antigens located on cell membrane, in the cytoplasm and in cell nuclei. This solution allows for significant increase of specific staining signal without producing non-specific background staining in specimens over-fixed with formalin as well as in long-stored archived tissues. To make a working solution dilute Antigen Retrieval Solution 10X concentrate to 1X concentration with distilled water. First deparafinize formalin-fixed paraffin-embedded tissue sections and then immerse them into Antigen Retrieval Solution heated between 95 and 100C for 10 minute, remove from heat and let Antigen retrieval Solution with immersed slides to cool down for 20-40 minutes. Pressure cooker or vegetable steamer can be used for retrieval procedure.
