| Notes | This gene encodes a cytokine that acts as a growth and differentiation factor for both B cells and eosinophils. The encoded cytokine plays a major role in the regulation of eosinophil formation, maturation, recruitment and survival. The increased production of this cytokine may be related to pathogenesis of eosinophil-dependent inflammatory diseases. This cytokine functions by binding to its receptor, which is a heterodimer, whose beta subunit is shared with the receptors for interleukine 3 (IL3) and colony stimulating factor 2 (CSF2/GM-CSF). This gene is located on chromosome 5 within a cytokine gene cluster which includes interleukin 4 (IL4), interleukin 13 (IL13), and CSF2 . This gene, IL4, and IL13 may be regulated coordinately by long-range regulatory elements spread over 120 kilobases on chromosome 5q31. |

| Protocol | ELISA: JES1-39D10 monoclonal antibody is useful as a capture or detection antibody in a sandwich ELISA for quantifying human IL-5 protein levels. 1-3,5 Purified JES1-39D10 antibody may be paired with biotinylated JES1-5A10 antibody (OASB01217) as the detection antibody, with purified recombinant human IL-5 as the standard. Or, conversely, biotinylated JES1-39D10 may be paired with purified JES1-5A10 (OASB00115) as the capture antibody. Immunofluorescence/Flow Cytometry: JES1-39D10 antibody is useful for intracytoplasmic staining and flow cytometric analysis to identify and enumerate IL-5-positive cells within mixed cell populations.5,6 Recombinant human IL-5, unlabeled JES1-39D10 (OASB01218), or Rat IgG1 (OASB00087or OASB00091) should be used as a specificity control. Western Blotting: The purified JES1-39D10 antibody is also useful for Western blotting applications. A concentration of 1-5ug/mL (in conjunction with AP-labeled Goat Anti-Rat-Ig, OASB01897) has been found to enable detection of <= 100 ng/lane of recombinant human IL-5 under reducing conditions. Since applications vary, each investigator should determine the optimal concentration appropriate for individual applications. |
