Additional Information | EHMT2 (G9a) is a lysine-preferring histone methyltransferase. EHMT2 can methylate itself at K165 and this automethylation is necessary and sufficient to mediate in vivo interaction of EHMT2 with heterochromatin protein-1 (HP1) (1). EHMT2 is responsible for the majority of dimethylation of histone H3 at lysine 9 (H3K9me2) and is required for the efficient repression of developmentally regulated genes during embryonic stem cell differentiation (2). EHMT2 is overexpressed in various cancers and can can specifically methylate p53 at Lys(373). EHMT2 polymorphisms have been associated with colorectal cancer. |

Protocol | 50 µl reaction mix (50 mM Tris pH 8.8, 5 mM MgCl<sub>2</sub>, 4 mM DTT, 20 µM S-adenosylmethionine and 0-40 ng G9a) is added to wells coated with the substrate. Incubate for 1 hour. Add antibody against methylated K9 residue of histone H3, incubate at room temperature for 1 hour. Finally, incubate 1 hour with the Europium-labeled secondary antibody. Time resolved fluorescence measured at room temperature (λexc=330 nm, λem=620 nm). |
